dc.description.abstractDengue is a wide-spread disease caused by dengue virus (DENV), a member of the Flaviviridae. Dengue causes various clinical manifestations, from dengue fever (DF) to severe forms of the disease, such as dengue shock syndrome (DSS) and dengue hemorrhagic fever (DHF). Trenggalek is one of the areas in East Java, Indonesia where the dengue outbreak is re-occurring. In 2016, Trenggalek had more than 1000 cases reported, one of the highest in the East Java region. Even though, such high dengue cases are constantly re-occurring in Indonesia yet the serotype and genetic profile of dengue cases are limited. Therefore, the aim of this study is to detect the presence of DENV in the collected samples and to determine the serotype and the genetic profile of the isolated virus. Collected blood serum from suspected patients were cultured in C6/36 and vero cell in minimum essential medium (MEM) for virus propagation. Pre-screening, enzyme-linked immunosorbent assay (ELISA) was used to detect the presents of the DENV antigen. The ELISA-positive samples were selected for the RNA extraction. Then PCR and sequencing were used to determine the serotype and genetic profile. Most of the samples showed negative result due to RNA integrity issue. The remaining 2 positive samples were confirmed to be DENV-4 strain. The genetic profile of the positive samples were identified to be in the same clade with South East Asian and Papua New Guinean isolate.en_US
