Abstract: This study aims to generate molecular infectious clones, incorporating reporter genes (GFP and SfCherry), for flaviviruses and noroviruses—namely, Kunjin, Zika, and Murine Norovirus (MNV). The objective is to use viral constructs for antiviral compound screening and unravel their mechanisms of action. Employing a diverse array of methods, including PCR, CPER, transfection, fluorescence, and diagnostic tests like Plaque Assay and Western Blot, the investigation successfully produced molecular infectious clones for the Kunjin Virus. Regrettably, the construction of molecular infectious clones for the Zika Virus encountered challenges. Conversely, Murine Norovirus (MNV) demonstrated success in transfection methods and Western Blot assays but exhibited no conclusive results in the Plaque Assay. These findings contribute valuable insights into virological studies, presenting both successes and areas for further exploration in refining antiviral research methodologies.
